In this study, the development of a toxicity evaluation method for dioxins in human milk by enzyme-linked immunosorbent assay (ELISA) was reported. A total of 17 human milk samples were tested by ELISA and by gas chromatography/mass spectrometry (GC/MS) to assess whether the ELISA performed on samples obtained from primiparas could be considered as reliable enough for identifying a dioxins contamination in human milk. The concept of toxicity equivalent quantity (TEQ) screening was validated by comparing TEQ values for a set of human milk samples to the ELISA responses predicted for those samples. A fairly good correlation (r=0.920) between immunoassay and GC/MS was achieved for human milk. This ELISA should be useful for biological samples monitoring. 相似文献
Polybrominated diphenyl ethers (PBDEs) were measured in 2004 in 105 breast milk samples collected from 13 regions of Japan (Hokkaido, Akita, Miyagi, Tokyo, Gifu, Fukui, Kyoto, Hyogo, Wakayama, Shimane, Yamaguchi, Kochi and Okinawa). Six congeners (BDE-28, BDE-47, BDE-99, BDE-100, BDE-153 and BDE-154) were determined by gas chromatography /mass spectrometry (GC/MS). Total PBDE levels ranged from 0.01 to 23.0 ng/g lipid (geometric mean (GM), 1.34 ng/g lipid). BDE-47 (GM, 0.66 ng/g lipid, 59% of sigmaPBDE) was the most abundant congener present in breast milk and was detected in 99% of the samples. Total PBDE levels were higher in northern Japan than in other regions. We analyzed the effects of occupation, age, smoking status, alcohol consumption and number of deliveries on total PBDE levels. None of these factors were significantly associated with the level of PBDEs. The present study revealed that the current level of exposure to PBDEs in Japan is lower than that in the USA or Sweden. GMs (ng/g lipid) (GSD, geometric standard deviation) and medians (ng/g lipid) of PBDE levels in each district are as follows: Hokkaido 2.70 (1.70), 2.74; Akita 4.49 (2.19), 5.44; Miyagi 1.77 (4.37), 1.11; Tokyo 1.39 (2.09), 1.63, Gifu 2.83 (4.79), 2.23; Fukui 1.05 (2.34), 1.18; Kyoto 1.31 (2.95), 1.33; Hyogo 1.02 (2.69), 0.88; Wakayama 1.33 (3.80), 1.70; Shimane 0.83 (2.51), 0.66; Yamaguchi 1.74 (2.82), 1.76; Kochi 0.50 (2.69), 0.74 and Okinawa 1.91 (2.75), 1.22. Because of the remarkable chemical structure of perfluoroalkyl and polyfluoroalkyl substances (PFAS), as well as the complex conditions of water, selecting an appropriate adsorbent for treating PFAS is critical. Adsorption needs to be environmentally friendly, low cost, and consider the types of adsorbents that work well in mixed PFAS solutions. In the present study, we used mixed PFAS to estimate the PFAS activity. This research aimed to evaluate and compare the efficacy of the adsorption of PFAS from water using different adsorbents: granular activated carbon (GAC), IRA 910 (strong anion resin), and DOWEX MB-50 (mixed exchange resin). Batch adsorption isotherms and kinetic studies were performed for perfluorooctanoic acid (PFOA), perfluorooctane sulfonic acid (PFOS), and perfluorohexane sulfonic acid (PFHxS). Freundlich models consistently described the kinetic behavior with a high correlation coefficient (R2 > 0.98). PFAS adsorption capacities on GAC and IRA910 were dependent on the chain length (PFOS > PFOA > PFHxS). The adsorption capacity of DOWEX MB-50 decreased because of the sulfonate effects (PFOS > PFHxS > PFOA). The rate constants (k2) that represented the adsorption of PFAS on different adsorbents observed within 96 h were accurately determined by the pseudo-second-order (PSO) model. GAC achieved followed the relationship k2(PFOS) > k2(PFOA) > k2(PFHxS). Furthermore, k2 of IRA910 decreased in the order of k2(PFOA) > k2(PFOS) > k2(PFHxS), implying that IRA910 promoted hydrophobicity more significantly on the adsorption of PFCAs than perfluoroalkane (-alkyl) sulfonic acids. The kinetics of DOWEX MB-50 revealed k2(PFHxS) > k2(PFOS) > k2(PFOA) because gel-type resins like DOWEX MB-50 are more suitable for shorter-chain PFAS. Recently, reclaimed asphalt pavement (RAP) with high RAP contents with rejuvenator agents have been implemented in Japan. However, a unified view of the durability of RAP mixtures with rejuvenators has not been established. Therefore, this paper aims to assess the durability of RAP mixtures blended with different types of rejuvenators through combined aging, compared with that of hot mixed asphalt (HMA) mixture. Mechanical and physical properties of RAP mixtures with rejuvenators subjected to a combination of moisture, heat, and oxidation were investigated. Saturation aging tensile stiffness conditioning was used to simulate the combined aging. Four types of RAP mixtures were prepared by employing two types of oil-based rejuvenators with two RAP contents. HMA mixtures were also prepared. The test results demonstrated that the RAP mixtures did not exhibit a reduction in the retained stiffness, whereas the HMA mixtures exhibited a significant drop in stiffness with conditioning. In addition, the RAP mixtures exhibited a longer residual service life than the HMA mixtures after conditioning. Detailed analyses revealed differences in the stiffness and fatigue behaviors of the RAP mixtures with different types of rejuvenators. Finally, certain aspects of the deterioration mechanisms of RAP mixtures with rejuvenators were discussed.
Material flow analysis (MFA) is an effective tool for waste management, but low- and middle-income countries lack essential data for MFA. This study proposed a simplified MFA (sMFA) utilizing local expert judgment (LEJ) and examining the impact of simplification on its uncertainty. A stochastic sMFA model was developed for nitrogen and phosphorus in urban Mandalay, Myanmar. This model was compared with the intensive MFA (iMFA) model employing intensive surveys for primary data collection. For the total loadings to the environment, the medians of the sMFA were higher by 3% and 11%, respectively, for nitrogen and phosphorus than those of the iMFA. The widths of the 80% confidence intervals of these loadings in the sMFA, normalized by those in the iMFA, were − 0.05 and − 0.11, respectively. The three largest flows to the environment were the same for the two models: on-site sanitation effluent/leakage, greywater, and industrial wastewater. Large median gaps between the models were observed for industrial wastewater, fecal sludge, and human excreta, associated with informal waste management, whereby LEJ did not work well. Overall, the sMFA demonstrated a good estimation of nitrogen and phosphorus flows with limited increase of uncertainty, still requiring focused attention on informal waste streams.
